山茶叶片可溶性蛋白双向电泳技术的建立 被引量:9

摘要 建立了山茶低温诱导蛋白研究中叶片蛋白双向电泳技术体系.采用固相pH胶条在IPGphor TM等电聚焦系统上进行等电聚焦,在Ettan TM DaltⅡ高通量电泳仪上进行SDS-PAGE电泳,以银染法染色,得到了分离效果良好的蛋白质双向电泳图谱。讨论了蛋白沉降方法、蛋白提取液的组成及其他技术环节对蛋白图谱的影响。 This paper reported the improvement in 2-D gel electrophoresis proteome of the cold-induced proteins of the leaf of Camelliajaponica. An IPGphor TM unit with immobile pH gradient strips was used in and the Ettan TM Dalt Ⅱ system in SDS-PAGE. The 2-D gel was stained by methods of silver, and the better separating protein pattern could be gained by this method. The effect of protein sedimentation method, composition of protein extraction buffer and other parameters on protein pattern were discussed.
