摘要
[目的]研究产色素温和气单胞菌的致病机理。[方法]从不同来源的病鳖和病鳗体内分离到4株产生棕色色素温和气单胞菌,并采用平板法进行检测。[结果]对鲫鱼胸基部注射9×108cfu/ml菌悬液0.4ml后,BA10、BA15和BA16株对鲫鱼的致死率在33.3%以上,M1株对鲫鱼无致死性。采用超声波破碎-SI法提取外膜蛋白(OMPs),SDS-PAGE电泳结果显示,4株温和气单胞菌的外膜蛋白相对分子质量范围在20.1~66.2kDa,其中1个鳗鱼分离株M1株属OMP1型,由8条主要蛋白带组成;其余3个鳖源分离株BA10、BA15和BA16株属OMP2型,由9条主要蛋白带组成。[结论]产色素与致病性无相关;4株产色素温和气单胞菌之间存在共同抗原;产色素温和气单胞菌的OMP型可能与菌株来源有关。
[Objective] The aim of the research was to study the pathogenic mechanism of A eromonas sobria producing pigment. [Method] Four strains ofA. sobria that produced brown pigment were isolated from different sources of diseased turtles and eels and determined by plate method. [Result] After 0.4ml 9×10^8cfu/ml bacterial suspension was injected into the basal chest of crucian, the lethality rate of strains BA10, BA15 and BA16 to crucian were all over 33.3%, but strain M1 had no lethality to crucian. Outer membrane proteins were extracted by ultrasonic waves breaking -SI method. The results of SDS-PAGE showed that the relative molecular weight of outer membrane proteins of 4 strains of A. sobria were in 20.1-66.2 kDa. Among them, one isolated strain M1 from crucian belonged Type OMP1 and it was consisted of 8 main protein bands. Other 3 isolated strains from turtle including BA10, BA15 and BA16 belonged to Type OMP2 and it was consisted of 9 main protein bands. [Conclusion] There is no correlation between pigment production and pathogenicity. There was common antigen among 4 strains ofA. sobria. Type OMP ofA. sobria producing pigment was possibly related with the strain source.
