摘要
[目的]研究蔗糖、聚乙二醇(PEG)、脱落酸(ABA)及活性炭(AC)对芸芥体胚成熟的影响。[方法]以青城芸芥的无菌苗为试验材料,通过观察诱导体胚的形态和调查成熟体胚及正常体胚的比例,研究各因素对芸芥体胚成熟的影响。[结果]高浓度蔗糖(609、0 g/L)或添加适当浓度的PEG或一定量活性炭的条件下,芸芥体胚成熟均较好,生长快,直径达2 mm左右。45 g/L PEG最适于诱导芸芥体细胞胚胎的成熟,体胚成熟率和正常体胚率分别达35.0%和53.1%。1 g/L AC的效果最好,体胚成熟率和正常体胚率分别达30.6%和46.0%。在添加ABA的条件下,芸芥体胚成熟率和正常体胚率都低于对照。[结论]高糖培养基、添加适当浓度的PEG和一定量的活性炭均有利于芸芥体胚的成熟,ABA不利于芸芥体胚的成熟。
[Objective] The purpose was to research the influences of the factors such as sucrose, polyethylene glycol (PEG), abscisic acid (ABA) and activated carbon (AC) on the maturation of Eruca sativa somatic embryos. [Method] With aseptic seedlings of Qingcheng E. sativa as tested material, the influences of various factors on the maturation of E. sativa somatic embryos were researched through observing the morphology of induced somatic em- bryos and investigating the proportions of mature and nomkal somatic embryos. [Result] Under the condition of high sucrose concn. (60 and 90 g/L), adding PEG at suitable concn, or adding a certain AC, the E. sativa somatic embryos matured better and grew fast, with diameter reaching about 2 mm. PEG at 45 g/L was most suitable for inducing E. sativa somatic embryos to mature, with matured and nomkal somatic embryos reaching 35.0% and 53.1% resp. The effect of AC at 1 g/L was best, with matured and nomkal somatic embryos reaching 30.6% and 46.0% resp. Under the condition of adding ABA, the rotes of matured and normal E. sativa somatic embryos were lower than that of CK. [ Conclusion] The medium with high sugar content, additions of PEG at suitable concn, and addition of a certain AC were in favor of the maturation of E. sativa somatic embryos. ABA went against the matumtion of E. sativa somatic embryos.
