中药甘遂注射液的毒性试验研究

Toxicological Studies on Injection of Euphorbia Kansui Liou

本文报道了雌性小鼠腹腔注射中药甘遂注射液,LD_(50)为88mg/kg,相当于人体实际用量的17.6倍;致突变试验为阴性结果,其中包括小鼠骨髓细胞染色体畸变分析和基因突变试验;用甘遂注射液引产后对小鼠再次受孕率无影响;在5mg/kg和1mg/kg剂量下均有明显胚胎毒性,但对存活胎仔无致畸作用。

The results of toxicological evaluation on injection of Euphorobia kansui Liou showed that it is an acute toxic drug, the LD_(50) for female mice was 88mg/kg which is 17.6 times the dose actually used by human being, Euphorbia kansui Liou has neither mutagenic action for body cells nor genotoxicity for Salmonella typhinurium strains . Also , there is no significant influence for repregnant rate of the mice that once aborted by using this injection . Under both 1 mg/kg and 5 mg/kg doses,. the absorbed rate of fetus increased while the living rate decreased for the pregnant rats, suggesting that injection of Euphorbia kansui Liou has some embryotoxicity, but no teratogenic effect was found for the survive fetus.
