细胞因子信号转导抑制因子3和5在儿童过敏性哮喘特异性免疫治疗中变化的研究 被引量:5

Change of SOCS3 and SOCS5 mRNA in specific immunotherapy for children with allergic asthma

摘要 目的观察在儿童过敏性哮喘特异性免疫治疗过程中,与TH1/TH2细胞分化相关的细胞因子信号转导抑制因子(SOCS)3和SOCS5的mRNA在外周血单个核细胞(PBMC)中表达水平的变化。方法于2006年4月至2007年10月对郑州大学第一附属医院收治的39例过敏性哮喘患儿,采集其特异性免疫治疗前、治疗15周和治疗51周的PBMC,提取总RNA,采用SYBRGreenⅠ逆转录荧光定量PCR的方法检测每组SOCS3和SOCS5mRNA的表达情况。结果随着特异性免疫治疗的进行,患儿SOCS3的表达逐渐降低,SOCS5的表达15周时稍有降低,51周时又增高,SOCS5/SOCS3的比率逐渐升高。结论SOCS3和SOCS5在儿童哮喘的特异性免疫治疗中可能起重要作用。 Objective To survey the change of suppressor of cytokines signaling-3(SOCS3)and suppressor of cytokine singnaling-5(SOCS5)mRNA in peripheral blood mononuclear cells(PBMCs)in specific immunotherapy(SIT)for patients with allergic asthma.Methods The patients with allergic asthma were divided into three groups:pre-SIT(0 week),post-SIT for 15 weeks and post-SIT for 51 weeks.The transcription factors of SOCS3 and SOCS5 mRNA in PBMCs were detected by SYBR GreenⅠreal-time RT-PCR.Results The ratio of SOCS5 to SOCS3 mRNA gradually increased with the progression of SIT.Conclusion SOCS3 and SOCS5 may play an important role in SIT for asthma and SIT.
