摘要
目的探讨免疫磁珠法分离CD34+细胞的方法。方法应用密度梯度离心法分离骨髓单一核细胞,后采用免疫磁珠法从单一核细胞中分选出CD34+细胞,流式细胞仪鉴定其纯度。结果各样品分离出的CD34+细胞占骨髓单一核细胞含量的(2.19±0.12)%,经流式细胞仪鉴定应用免疫磁珠分离的CD34+细胞纯度为(94.6±2.1)%。结论免疫磁珠法是分离CD34+细胞较理想的方法。
Objective To explore a method for isolating CD34^+ cells from bone marrow. Methods Density gradient centrifngation and immunomagnetic bead method were used to isolate bone marrow derived mononuclear cells and CD34^ + cells, respectively. The purity of CD34 ^+ cells was determined by FACS method. Results The percentage of CD34 ^+ cells isolated was(2.19 ± 0.12) % in bone marrow derived mononuclear cells using immunomagnetic bead method, and the purity of CD34 ^+ cells was (94.6 ± 2.1) % by FACS method. Conclusion Immunomagnetic bead method is an ideal way to isolate CD34^+ cells from bone marrow.
