摘要
目的建立流式细胞术计数外周血嗜碱性粒细胞的实验方法。方法采用流式细胞术,根据嗜碱性粒细胞表面抗原的特性Lin1-,CD123+,HLA-DR-,计数了30例健康体检者及10例慢粒急变患者外周血嗜碱性粒细胞,同时用血细胞分析仪,人工涂片计数嗜碱性粒细胞。结果流式细胞术计数方法在低值(0.26%)和高值(4.27%)的精密度分别为CV=5.3%(n=10)和CV=4.1%(n=10)。在30例健康体检者中,流式细胞仪法与自动血细胞分析仪法计数嗜碱性粒细胞结果有显著差异,(P<0.01,r=0.003)。10例慢粒急变患者流式细胞仪计数嗜碱性粒细胞与人工血涂片分类比较无显著差异且相关良好,(P>0.05,r=0.918)。结论流式细胞仪法计数外周血嗜碱性粒细胞结果准确,可作为标准的计数嗜碱性粒细胞方法。
Objective To establish flowcytometric method for counting basophiles in peripheral blood. Methods Basophiles were measured in whole blood by flow cytometry after staining with antibodies composite of Lin,HLA-DR and CD123. Enumerated the percentages of basophiles in 30 healthy adults and 10 patients with chronic myelocytic leukaemia by flow cytometry,automatic hematology analyzer and microscope, respectively. Results The experimental variability of assay for high data and low data were 4.1% and 5.3 % respectively. The percentages of basophiles in 10 CML patiens were excellent correlation between the flowcytometric method and manual counts by microscope (P〉0.05,r=0.918). There were no correlation between the flowcytometric method and automatic hematology analyzer for counting basophiles in heaithy adults.Conclusion Provide a novel and rapid method for the flowcytometric enumeration of basophiles in whole blood.
