塞来昔布对大鼠癫痫持续状态海马神经元凋亡及Bcl-2表达的影响 被引量:1

摘要 目的探讨塞来昔布对大鼠癫痫持续状态(SE)后海马神经元凋亡的影响及其可能机制。方法采用氯化锂-匹罗卡品制作大鼠癫痫持续状态模型。Wistar大鼠54只,随机分为模型组、塞来昔布组和对照组。采用末端脱氧核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL)观察SE后12h、24h、72h和7d海马神经细胞凋亡的动态变化,免疫组化法观察各组海马Bcl-2的表达。结果对照组未见TUNEL阳性细胞;模型组SE后12h可见较多TUNEL阳性细胞,72h达高峰,7d减少;塞来昔布组各时间点凋亡细胞数均明显少于模型组(P<0.01)。对照组Bcl-2有基础量的表达;模型组SE后12h观察到Bcl-2表达增多,24h减弱,72h后仅有少量表达;塞来昔布组24h后各时间点Bcl-2表达均高于模型组(P<0.01)。结论塞来昔布可增加海马Bcl-2的表达,对癫痫发作后神经元凋亡具有保护作用。
