eIF4E Expression and Its Role in Malignancies

摘要 真核细胞翻译起始因子4E(eIF4E)是最重要的翻译起始因子,在真核细胞蛋白质合成中起重要作用。近年来研究显示,eIF4E过表达与肿瘤的发生发展关系密切。与正常组织及良性肿瘤相比较,发现很多恶性肿瘤和肿瘤旁组织中eIF4E过度表达,并与肿瘤的侵袭转移能力呈正相关。研究发现,eIF4E通过多方面调控恶性肿瘤相关mRNAs的翻译,包含细胞有丝分裂过程、激活原癌基因、血管形成、增强自分泌、细胞存活、侵袭及与细胞外环境的交通。eIF4E与肿瘤的密切关系为临床治疗肿瘤提供了新思路,更有可能成为肿瘤治疗的共同靶点和肿瘤发展与预后的预测因子。 eIF4E is an important eukaryotic translation initiation factor, eIF4E overexpression has been demonstrated in many human tumors and to be related to disease progression. A deeper understanding of the roles of eIF4E in malignancy will improve the capacity to exploit eIF4E as a therapeutic target and as a marker for human cancer progression.
