摘要 目的蛋白酶激活受体(PARs),为G蛋白偶联受体(GPCR)家族中的新成员,参与过敏反应的发生,粒-巨噬细胞集落刺激因子(GM-CSF)具有调节免疫应答的功能,然而GM-CSF对肥大细胞PARs表达的影响还不甚清楚。因此本研究探讨GM-CSF对肥大细胞P815 PARs表达的影响情况。方法肥大细胞P815激发后,收集各时间点的细胞。细胞处理后采用实时定量PCR和流式细胞术的方法在信使核糖核酸(mRNA)和蛋白水平检测PAR-1、PAR-2、PAR-3和PAR-4表达情况。结果GM-CSF上调PAR-2蛋白和mRNA在肥大细胞P815上的表达;GM-CSF上调PAR-1和PAR-3 mRNA在肥大细胞P815的表达;GM-CSF抗体的应用可以阻断GM-CSF对PARs蛋白表达的调节作用。结论GM-CSF能够调节肥大细胞PARs表达,二者可能在过敏反应和炎症中存在更加复杂的相互作用。 Objective Protease activated receptors ( PARs), a new subfamily of G protein - coupled receptors, are involved in the pathogenesis of allergic diseases, and GM - CSF plays a role in adaptive immune response. However, little is known of the effect of GM - CSF on the modulation of mast cell pretease activated receptors expression. In the present study, we examined potential influence of GM - CSF on mast cell PAR expression. Methods After being challenged, some of cells were treated and analyzed by real time RT - PCR to detect expression of PARs at mRNA level, and some of P815 cells were treated and analyzed by fiowcytometry to detect expression of PARs at protein level. Results We demonstrated that GM - CSF upregulated mast cell PAR2 expression both at mRNA level and at protein level. GM - CSF upregulated PAR1 and PAR3 expressions at mRNA level. Modulation of expression of PARs at protein level was blocked by administration of GM - CSF blocking antibody. Conclusion Given the supporting data outlined above, it would seem that GM -CSF can modulate mast cell proteinase activated receptors expression, and both of them would have important role in the immune response and inflammation.
