摘要
目的:探讨二十二碳六烯酸(DHA)-磷脂的抗肿瘤和免疫调节作用。方法:从鸢乌贼卵(Symplectoteuthis oual-aniensis)中提取制备DHA-磷脂,并制备单室脂质体;采用不同剂量的DHA-磷脂脂质体灌胃移植性肿瘤模型小鼠,分别测定其瘤重、脾脏指数、胸腺指数、血清溶血素含量、足跖增厚值和巨噬细胞吞噬能力。结果:DHA-磷脂脂质体能显著抑制小鼠S180肉瘤的生长,其平均抑瘤率为36.87%,提高荷瘤小鼠脾脏脂数(P<0.05),显著提高血清溶血素含量(P<0.05,P<0.01),提高迟发型变态反应水平(P<0.05,P<0.01),促进巨噬细胞吞噬能力(P<0.01)。结论:DHA-磷脂脂质体能全面调节机体的特异性和非特异性免疫功能,抑制肿瘤细胞的生长。
Aim: To investigate the anti-tumor activities and immune regulation of the docosahexaenoic acid (DHA) -phosphatidylcholine (PC) liposomes in S180-bearing mice. Methods: DHA-PC (containing 30% DHA) separated from cuttlefish spawns was used to prepare unilayer liposomes. Different doses of DHA-PC liposomes were given to the transplanted tumor-bearing mice, and the tumor weight, spleen index, thymus index, hemolysin content, delayed type hypersensitivity and the phagocytosis ability of peritoneal macrophage were measured. Hesuits: It was shown that administration of DHA-PC liposomes inhibited the growth of tumor in S180-bearing mice and that the average tumor-inhibitory rate was 36.87%. Furthermore, there were increases of the spleen index (P 〈 0.05), the hemolysin content ( P 〈 0.05, P 〈 0.01), delayed allergic level( P 〈 0.05, P 〈 0.01), and phagocytosis capability of peritoneal macrophage( P 〈 0.01). Conclusion: DHA-PC liposomes could regulate both the spe-cific and non-specific immune functions, and inhibit the growth of tumor cells.
