摘要
目的:观察血管紧张素Ⅱ(AngⅡ)对成人脂肪间充质干细胞(ADMSCs)向心肌细胞方向分化的诱导作用,并与5-氮杂胞苷(5-aza)作比较.方法:自成人脂肪组织中分离培养ADMSCs,分别用AngⅡ(Ⅰ组)和5-aza(Ⅱ组)诱导ADM-SCs,同时设立对照组(Ⅲ组),光镜下观察细胞的生长情况及形态学变化,电镜下观察细胞的超微结构,免疫细胞化学方法检测心肌特异性肌钙蛋白-Ⅰ(cTn-Ⅰ),MTT法检测细胞活性,流式细胞术分析细胞周期及凋亡.结果:Ⅰ,Ⅱ组ADM-SCs诱导后呈现类似心肌细胞的形态学特征,第28日Ⅰ,Ⅱ组均表达cTn-Ⅰ,两组间诱导转化率无差别,其中Ⅰ组ADM-SCs增殖迅速,较少凋亡细胞.结论:AngⅡ在促进ADMSCs增殖的同时诱导其向心肌细胞分化,优于传统的化学诱导剂5-aza.
AIM: To study the induction effect of angiotensin Ⅱ (Ang Ⅱ ) on the differentiation of adult adipose-derived mesenchymal stem cells (ADMSCs) into myocardial cells, and to compare its effect with 5-azacytidine (5-aza). METHODS: ADM- SCs were isolated and cultured from adult adipose tissue. Ang Ⅱ ( Group I ) and 5-oza ( Group Ⅱ ) were added to induce ADMSCs, while control group ( Group Ⅲ ) was set up. Cell growth and morphological changes were observed with light microscope. Electron microscope was used to observe cell ultrastructure, while immunocytochemical methods was adopted to test cardiac-specific troponin- Ⅰ (cTn- Ⅰ ). MTT was used to assay cell viability and flow cytometry was to analyze cell cycle and apoptosis. RESULTS : ADMSCs in Group I and Group II showed similar myocardial cells in morphological features. Twenty-eight days after induction, ADMSCs in Group Ⅰ and Group Ⅱ both expressed cTn- Ⅰ, and induced conversion rate in these 2 groups had no discrimination. ADMSCs in Group I had higher proliferation rate and lower apoptosis rate than those of Group Ⅱ. CONCLUSION : Ang Ⅱ can not only promote ADMSCs proliferation but also induce differentiation to the direction of myocardial cells. Its effects is better than that of traditional chemical inducer 5-aza.
