苯乙酸对胶质瘤C6细胞中c-myc和Caspase-3表达的影响

摘要 目的观察诱导分化剂苯乙酸(phenylacetate,PA)对c-myc和Caspase-3表达的影响,探讨PA诱导胶质瘤C6细胞凋亡的机制。方法体外培养胶质瘤C6细胞,实验组给予PA浓度5.0mmol/L诱导72h,对照组不用PA,分别用免疫细胞化学检测c-myc和Caspase-3的表达,图像分析比较表达水平的差异。结果免疫细胞化学检测,两对照组和实验组中c-myc的表达分别为0.214±0.013和0.153±0.007,Caspase-3的表达分别为0.149±0.004和0.206±0.033。结论PA能够下调c-myc的表达和增强Caspase-3的表达,其诱导胶质瘤C6细胞凋亡的机制可能与此有关。 Objective To study the effects of differentiation inducer phenylacetate(PA) on the expression of c-myc and Caspase-3 in rat C6 glioma cells and explore the mechanism of PA action.Methods C6 glioma cells were cultured in vitro.In experiment group,C6 cells were induced by PA at concentration of 5.0mmol/L for 72 hours.In control group,no PA was administrated.The expression of c-myc and Caspase-3 was detected by using an immunocytochemistry technique.The differernces of the imaging presentation in the two groups were analyzed.Results The expressions of c-myc in control group and experiment group were 0.214±0.013 and 0.153±0.007,respectively,while the expression of Caspase-3 were 0.149±0.004 and 0.206±0.033,respectively.Conclusion PA can down-regulate the expression of c-myc and enhance the expression of Caspase-3,which might consequently induce apoptosis in C6 glioma cells.
