摘要 目的:考察AB-8树脂对珍珠菜提取液中总黄酮的吸附、解吸附性能及影响因素。方法:采用紫外分光光度法测定总黄酮的含量,分别考察了样品液浓度、pH值和流速对AB-8树脂吸附珍珠菜总黄酮的影响,以及洗脱剂乙醇浓度和流速对解吸附效果的影响。结果:样品液总黄酮浓度10 mg/mL、pH值5.07、流速1.0 mL/min时,AB-8树脂对珍珠菜总黄酮的吸附效果较好;洗脱剂为70%乙醇,流速1.5 mL/min时,洗脱完全,固体总黄酮含量较高。结论:在上述条件下,AB-8树脂可用于珍珠菜提取液中总黄酮的分离纯化。 OBJECTIVE To investigate the adsorption and desorption properties, and factors of AB - 8 res- in for total flavones in Lysimachia clethroide Duby. METHODS UV speetrophotometry was used to determine the concentration of total flavones. Factors of concentration, pH and fluid velocity of samples that affect adsorption of flavones, and concentration and fluid velocity of ethanol that affect desorption of flavones were investigated. RESULTS The optimum conditions for AB - 8 resin to separate and purify the total flavones from Lysimachia clethroide Duby were as follows : concentration of total flavones of liquid samples was 10 mg/mL, pH was 5.07, and fluid velocity was 1.0 mL/min, concentration and fluid velocity of ethanol as eluant were 70% and 1.5 mL/min, respectively. CONCLUSION Total flavones in Lysimaehia elethroide Duby would be well separated and purified when AB -8 resin was used.
