摘要
目的:研究皂荚提取物对人肝癌细胞的增殖、相关癌基因表达的调控作用及端粒酶的影响。方法:体外培养人肝癌细胞bel-7402,检测不同浓度皂荚提取物(0.05mg/ml、0.1mg/ml、0.2mg/ml)对人肝癌细胞bel-7402细胞增殖、细胞周期、肝癌相关基因(bax、bcl-2、p53)的表达及端粒酶活性的影响。结果:0.05mg/ml~0.2mg/ml浓度组皂荚提取液对人肝癌细胞bel-7402细胞增殖、调控癌基因与抑癌基因、促进癌细胞凋亡、抑制端粒酶活性差异均具有显著性意义(P<0.05或P<0.01)。结论:皂荚提取物能够抑制人肝癌细胞增殖,促进细胞凋亡。
Objective: To study the influence Glelitsia sinensis Lam extractive on the proliferation, the regulation of cancerous gene and telomerase activity of human hepatocarcinoma cell. Methods: Bel-7402 cell was cultured in vitro and treater with different concentration of Glelitsia sinensis Lam (0. 05mg/ml, 0. lmg/ml, 0. 2mg/ml) . MTT was explored to detect the praliferation of bel-7402 cells. Flow cytometry was used to measure apptosis and cellcycle. RT-PCR and immunohistochemical method was adapted to assay the expressionef cancerousgene. Results: The coneentration of Glelitsia sinensis Lam from 0. 05mg/ ml to 0. 2mg/ml could manedkly inhibit the praliferation, the expression of cancerous gene and the thlomerase activity ( P 〈 0. 01 ) . Conclusion : The extractive of Glelitsia sinensis Lam can control the proliferation, accelerate the apoptotsis of bel-7402 cell.
