摘要
目的探讨更年春方对卵巢切除大鼠学习记忆的影响。方法10b12月龄SD雌性大鼠切除卵巢建立学习和记忆能力受损的模型,分别用更年春方、尼尔雌醇灌药12周后Morris水迷宫测定各组大鼠学习记忆能力。结果大鼠切除卵巢后,血清雌二醇(E2)水平和Morris水迷宫测定的学习和记忆能力显著下降。尼尔雌醇和高剂量更年春方可以改善大鼠切除卵巢后的学习记忆能力下降的症状,而对血清E2无明显影响。结论更年春方对卵巢切除导致的学习和记忆能力下降有改善作用,高剂量效果显著,其效果与尼尔雌醇效果相似。更年春方不提高卵巢切除大鼠血清E2水平。
Purpose To evaluate the effect of Gengnianchun formula (GNC) on learning and memory in ovariectomized (OVX) Sprague Dawley (SD) rats. Methods Ten- to twelve-month-old SD rats were randomized into mimic sham operation group(S), ovariectomized OVX group (N), OVX + high- dose GNC group (H), OVX + low-dose GNC group (L) and OVX + nilestriol group (E). A little fat around ovary was cut in S group rats. Ovariectomy was performed in four later groups. These rats were fed with NS or GNC or nilestriol for 12 weeks respectively. After 12 weeks, learning and memory were tested by Morris water maze. On the following day of Morris water maze test, blood was taken from the carotid artery of rats. The serum level of 17[3-estrodiol (E2) was measured. Results Twelve weeks after ovariectomy, serum E2 level and learning-and-memory in Morris water maze of N group rats were markedly decreased compared with rats in S group (P〈0.01 or P〈0.05). After treatment with high-dose GNC and nilestriol learning and memory was improved markedly (P〈0.01 or P〈0.05), but no significant change was observed in serum E2 level (P〉0.05). Conclusions High-dose GNC can ameliorate the reduction of learning-and-memory in ovariectomized rats, the effectiveness was similar to that of nilestriol. GNC can not improve serum E2 in OVX rats.
