摘要
目的建立测定人血浆中左氧氟沙星浓度的反相高效液相色谱荧光法(HPLC-FLD)。方法采用HPLC- FLD法测定,以Kromasil C_(18)柱为分析柱,0.4%三乙胺溶液(pH3.00)-乙腈(87:13,v/v)为流动相,流速为0.8 ml/min,激发波长为295 nm,发射波长为490 nm,洛美沙星为内标物。结果左氧氟沙星在0.0156~16.0μg/ml检测浓度范围内呈良好线性关系(r=0.9998),平均回收率为(100.1±2.5)%;日内差异≤3.9%,日间差异≤3.6%。结论该方法操作简便、灵敏、准确,适用于临床左氧氟沙星血浆浓度的监测。
Objective To establish a RP-HPLC method for determination of levofloxacin in Human plasma. Methods The HPLC-FLD method used Kromasil C18 (4.6 mm×150 ram,5 μm) column as an analytical column,a mobile phase consisted of a triethylamine-buffer(0.4%,pH3.00)and acetonitrile (87:13,v/v), at a flow rate of 0.8 ml/min,and the excitation and emission wavelengths were 295 nm and 490 nm respectively. Lomefloxacin was used as internal standard. Results The assay was linear over the concentration range of 0.0156-16.0 txg/ml (r=0.9998). The average recovery was (100.12±2.45)%. Intra-day and inter-day precision showed coefficients of variation within 3.9%(n=5)and 3.6%(n=5)over the four concentration levels. Conclusion This method is accurate ,sensitive and simple,and can be used to monitor the concentration of levofloxacin in the patients plasma.
