摘要
目的:建立黄芩的毛细管电泳指纹图谱(CEFP)并测定黄芩苷含量。方法:以50 mmol/L硼砂溶液(含5%乙腈,pH9.30)为背景电解质,石英毛细管(75 cm×75μm i.d.),有效长度63 cm,运行电压12 kV,紫外检测波长280 nm,重力进样15 s(高度10 cm)。用定性相似度SF、含量相似度Q、宏观含量相似度R等指数评价黄芩CEFP,以CZE测定黄芩苷含量。结果:以萘普生峰为参照物峰,确定了8个共有峰,10个产地黄芩的CEFP与黄芩苷的CEFP具有良好相似性。含量测定结果表明不同产地黄芩中黄芩苷含量有明显差异。结论:所建立的CEFP及含量分析方法均具有较好的重现性,两者结合为黄芩药材质量控制提供了新方法。
AIM: Capillary electrophoresis fingerprints (CEFP) and quantitative analysis methods of Radix Scutellariae were established. METHODS: All experiments were carried out in an uncoated fused silica capillary (75 cm×75 μm i. d. , effective length 63 cm) with a 50 mmol/L sodium borate solution (contained 5% acetonitrile, pH 9.30) under 12 kV while the detection wavelength was set at 280 nm and naproxen was selected as the internal standard. RESULTS: 8 co-possessing peaks were selected in the CEFP taking naproxen referential peak. The similarities between each of the ten places and the referential CEFP of Radix Scutellariae were evaluated both the qualitative similarity SF and the quantitative similariy Q, the apparent quantitative similarity R. CONCLUSION: The method of the CEFP and quantitative analysis are rapid, simple and accurate with a good repeatability and can be used for the quality control of Radix Scutellariae.
