摘要
采用响应面方法,对能够高效降解胆固醇的马红球菌F21-1的发酵培养基进行优化。在前期比较试验已筛选出碳源、氮源及表面活性剂最佳组合的基础上,通过Box-Behnken设计法对这3个因素进行响应面分析,以胆固醇降解率为响应值作响应面和等高线图。结果表明,马红球菌F21-1降解胆固醇发酵培养基添加物最佳成分配比为:胆固醇为1.49 g/L,蛋白胨为7.91 g/L,Tween-80为2.82 mL/L。培养基成分优化后,马红球菌F21-1菌株胆固醇降解率(87.80%)提高了22.53%。
The cholesterol-degrading fermentation medium of Rhodococcus sp. was optimized by using the response surface methodology. Firstly, the influences of different carbon, nitrogen sources and the surface active agent on cholesterol degrading rate were studied, and then base on this, the three factors were designed by Box-Behnken design and analyzed by Response Surface Analysis method. The response surface and the contour were graphed using cholesterol degrading rate as the response value. The result indicated that the best match of the ferment medium was: cholesterol 1.49 g/L, peptone 7.91 g/L, Tween-80 2.82 mL/L. After optimizing, cholesterol degrading rate (87. 80%) of Rhodococcus sp. F21-1 has been improved by 22.53%.
