Colorimetric Detection of Point Mutation Using Gold Nanoparticles and Double-stranded Probes

摘要 将链置换的高度特异性与纳米金凝聚变色的光学特性相结合,设计了一种新型的单碱基突变比色检测方法。本方法直接采用纳米金作为比色报告基团,以两个末端均带有巯基的双链DNA为特异捕获探针,利用互补序列和单碱基突变序列对双链探针置换能力的差异,实现了对单碱基突变的检测。本检测方法直观、快速、简便、成本低,pmol级的样品无需仪器就可以观察到颜色的变化。 A novel colorimetric point mutation detection method was developed by combininng the distancedependent optical properties of gold nanoparticles with the high specificity of strand displacement. Unmodified gold nanoparticles were used as reported groups. The probes consist of two complementary oligonucleotides of different length labeled with double thiol group. A perfectly matched target and single base mutation target can completely discrimination taking advantage of their different ability to displace double-stranded probes. The color of gold nanoparticle solution containing perfectly matched target and probes is red and the color of gold nanoparticle solution containing single base mutation target and probes is blue. The red color indicates that the double-stranded probes have been displaced. This new method is rapid, simple, low cost, and 1 pmol of target produces color changes observable without instrumentation.
