摘要
目的:研究神经生长因子(NGF)及其受体P75NGFR和TrkA在胰腺癌组织中的表达及其与临床病理学指标间的关系。方法:采用免疫组化技术检测NGF、P75NGFR和TrkA在65例胰腺癌、18例癌旁正常胰腺及慢性胰腺炎组织中的表达和分布,并分析其表达与临床病理学指标间的关系。结果:NGF及TrkA在胰腺癌组织中的表达较癌旁正常胰腺及慢性胰腺炎组织高;而P75NGFR的表达无显著性差异。NGF的表达与肿瘤大小密切相关.与淋巴结转移也有关;而P75NGFR和Tr—kA两者的表达只与淋巴结转移密切相关。NGF的表达与胰腺癌组织学分型、肿瘤分期、肿瘤大小、淋巴结转移都有关,而与肿瘤大小的关系最为密切;P75NGFR的表达仅与淋巴结转移有关;而TrkA的表达与淋巴结转移和肿瘤分期有关。淋巴结转移和TrkA的阳性表达与预后密切相关。结论:NGF、P75NGFR和TrkA在胰腺癌的发展中有一定的作用,TrkA与胰腺癌的关系尤为密切,可作为重要的靶标分子用于胰腺癌的基因治疗。
Objective:To explore the expression of nerve growth factor (NGF), its receptor P75NGFR and TrkA in the human pancreatic cancer tissues, and to study the relationship between the expression and clinical-pathological indices. Methods: The expression and morphological distribution of NGF, P75NGFR and TrkA were investgated in 65 pancreatic cancer tissues and 18 normal pancreas and chronic pancreatitis tissues by means of immunohistochemistry; the relationship between the expression and clinical-pathological indices was also investigated. Results: The expression of NGF and TrkA in human pancreatic cancer tissues was higher than that in the normal pancreas and chronic pancreatitis. Expression of P75NGFR had no significant change in the two groups. The expression of NGF was positively correlated with tumor size and was also associated with lymphatic invasion. The expression of P75NGFR and TrkA was closely correlated with lymphatic invasion. We also found that the expression of NGF was correlated with histological differentiation, tumor stage, tumor size and lymphatic invasion; the closest relationship was found between the expression of NGF and tumor size. The expression of P75NGFR was correlated with lymphatic invasion, the expression of TrkA was correlated with both lymphatic invasion and tumor TNM stage. Patient's prognosis after surgery was mainly correlated with lymphatic invasion and the positive expression of TrkA. Conclusion tNGF, P75 NGFR and TrkA participate in the progress of pancreatic cancer. TrkA is closely related to pancreatic cancer and can serve as an important target for gene therapy of pancreatic cancer in the future.
