摘要
制备离体大鼠伏核脑片,用细胞外记录法观察乙醇对谷氨酸钠诱发伏核神经元放电活动的影响,观察给药后5~15分钟内神经元放电频率的改变。结果显示:灌流谷氨酸钠后10分钟神经元放电频率显著增多(P<0.01),此反应可被乙醇抑制。说明乙醇对伏核神经元抑制作用的机制可能与谷氨酸介导的突触传递受抑制有关。
The nucleus accumbens septi(NAcc) is a key brain region in the rewarding effects of addictive drugs such as opiates and ethanol. The purpose of this study is to investigate ethanol actions on electrical activity evoked by glutamate in slice preparation of NAcc. Using extracellular recording, we found that the firing of NAcc neuron can be increased in 10~15 min by superfusing of 0.5 mmol·L -1 glutamate ( P <0.05), and this increasing responses can be inhibited by superfusing 0.2 μmol·L -1 ethanol in 5~10 min( P <0.01). The results suggest that a major effect of ethanol in NAcc is to reduce synaptic transmission mediated by glutamate.
