摘要
探讨精氨酸-甘氨酸-天门冬氨酸-丝氨酸(Arg-Gly-Asp-Ser RGDS)4肽诱导人结肠癌细胞SW480凋亡的作用。采用DAPI染色、DNA琼脂糖凝胶电泳以及流式细胞技术分析RGDS对人结肠癌细胞SW480凋亡的影响。结果表明,与对照组相比,RGDS处理细胞后,形态学观察显示DAPI染色的细胞核染色质凝聚、细胞溶解形成凋亡小体;DNA琼脂糖凝胶电泳出现典型的DNA梯状带(DNA ladder);流式细胞分析可见凋亡峰,凋亡率约是对照组的4.5倍。因此,RGDS具有诱导人结肠癌细胞SW480凋亡的作用。
To observe the apoptosis in human colon carcinoma SW480 cells exposed to RGDS (Arg-Gly- Asp-Ser), apoptosis was detected by DAPI staining, DNA agarose gel electrophoresis technique and flow cytometry. When cells were exposed to RGDS, the morphologic changes in DAPI-stained cell nuclei showed condensed or fragmented nuclei. A regularly spaced "ladder" shape of genomic DNA electrophoresis and obvious sub-Gl phase peak occurring in flow cytometry were observed for SW480 cells exposed to RGDS. The apoptotic portion of cells exposed to RGDS was 4. 5 folds increased, compared to the control cells. Therefore,RGDS was able to induce apoptosis in human colon carcinoma SW480 cells.
