羊膜移植后碱烧伤兔眼角膜PEDF和VEGF的表达

Expression of VEGF and PEDF in rabbits with corneal alkali burn following amniotic membrane transplantation

摘要 目的探讨羊膜移植对兔眼碱烧伤角膜色素上皮衍生因子(PEDF)和血管内皮生长因子(VEGF)表达的影响。方法新西兰白兔角膜碱烧伤模型被建立,左眼行羊膜移植,右眼作为对照。照相法观察兔眼碱烧伤角膜CNV及免疫印迹法检测不同时间点碱烧伤兔眼角膜中PEDF和VEGF的表达水平。结果羊膜移植组与对照组比较:羊膜移植组兔眼角膜碱烧伤后新生血管的生长速度较对照组慢(P<0.05);羊膜移植组兔眼角膜PEDF的表达水平较对照组高,而VEGF的表达水平较对照组低。结论羊膜移植上调PEDF的表达、下调VEGF的表达抑制兔眼角膜碱烧伤后CNV的形成。 Objective To examine the impact of amniotic membrane transplantation on pigment epithelium derived factor (PEDF) expression in rabbits with corneal alkali bum with regard to vascular endothelial growth factor(VEGF) expression. Methods The models with corneal alkali burn were established in New Zealand rabbits. Their left eyes were transplanted with amniotic membrane, and right eyes served as controls. Corneal PEDF, VEGF expression and corneal revasctdarization (CNV) were measured by Western blot and slit-lamp microscopy, respectively. Results Specimens without amniotic membrane transplantation disclosed the bigger CNV areas, enhanced VEGF, and reduced PEDF expression at different time points in rabbits with corneal alkali burn. In contrast, Specimens with amniotic membrane transplantation disclosed the smaller CVN areas, reduced VEGF, and enhanced PEDF expression at different time points in rabbits with corneal alkali burn. Conclusion Amniotic membrane transplantation inhibits the formation of CNV in rabbits with corneal alk ali burn. The effectiveness of amniotic membrane transplantation, however, seems to be jeopardized by a effect initiated by an enhanced PEDF and reduced VEGF expression.
