摘要
为建立定量测定鸡食用组织中喹烯酮的高效液相色谱检测方法,并依据测定的结果制定最高残留限量,确定其休药期。选用63只鸡,按正常饲料添加喹烯酮(75 mg/kg)饲喂2.5月后,按既定时间点采样。组织样品用乙酸乙酯等试剂提取处理纯化后进样,在312 nm波长处检测,流动相为甲醇-水(70∶30),流速为1.0 mL/min。结果表明,在试验条件下,脂肪、肝脏、肾脏、肌肉组织中的最低检出浓度均为0.005μg/g。平均回收率肌肉为99.23%±2.75%,脂肪为105.15%±8.65%;肝脏为93.55%±11.16%;肾脏为104.10%±9.80%。喹烯酮在肌肉皮脂中1 d后均无残留,而在肝脏和肾脏中的残留量也很小,2 d以后再无残留。精密度考察喹烯酮的日内和日间各组织各浓度的相对标准差均小于8.5%。因此,本试验测定方法灵敏度高,重复性好。停药后4 h所有可食用鸡组织中喹烯酮浓度均低于计算所得的安全组织浓度,故该药在鸡上使用无休药期,即休药期为0 d。
HPLC quantitative assay of quinocetone in edible chicken tissues was established to determine the withdrawal time. 63 chicken were given 75 mg/kg quinocetone additive in feed for two and half months and killed to collect tissue samples. After tissue samples were extracted and purified with actoactate and other reagents, they were measured at 312 nm wave length, methOH-water(70:30) mobile phase and 1.0 mL/ min flow rate. The result showed that the lowest concentrations measured in fat, liver, kidney and muscle tissues were all 0. 005 μg/g. The average recovery rates in muscle, fat, liver and kidney were (99.23 ± 2.75)%, (105.15±8.65)%, (93.55±11.16)% and (104.10±9.80)%, respectively. RSDs of quinocetone concentration in each tissue for intraday and interday were less than 8.5 %. quinocetone in muscle and fat was not existed for 1 day, and in liver and kidney for 2 days. As a result, the assay was sensitive and high repeatable. The withdrawal time of quinocetone in chicken was 0 day.
