摘要
目的:研究皂角刺皂苷(Saponins of spina gleditsiae,SSG)对前列腺癌细胞增殖及凋亡的影响。方法:实验分成不同浓度的给药处理组(SSG)、未加药的阴性对照组(NP)、以培养液为参照的空白对照组(CP)。对数生长期PC-3细胞用EDTA-胰蛋白酶消化后,接种于96孔培养板。贴壁后分别加入不同浓度的SSG溶液,每孔100μL,每组设6复孔。体外培养前列腺癌PC-3细胞给药处理48h后,MTT比色法检测前列腺癌PC-3细胞抑制率,流式细胞仪检测SSG对PC-3细胞凋亡率的影响。结果:与NP相比,SSG可显著抑制前列腺癌PC-3细胞的增殖(P<0.05);SSG50mg/L组、100mg/L组、200mg/L组的细胞凋亡率分别为6.91%,10.52%和16.50%,3组的细胞凋亡率均高于0mg/L组(P<0.05)。结论:SSG对前列腺癌PC-3细胞具有抑制增殖和诱导凋亡的作用。
Objective: To investigate the effects of Saponins of spina gleditsiae(SSG) on the proliferation and apoptosis in PC-3 cell. Methods: The experiment included treatment group (SSG),negative control group (NP),and control group (CP). After PC-3 cell were treated with various concentrations of SSG for 48 hours, PC-3 cell proliferation was determined by MTT colorimetric assay. The apoptosis rate of PC-3 cells were examined by flow cytometer. Results: The proliferation of PC-3 cells was significantly inhibited by SSG (P 〈 0.05 ). Compared with the negative group (NP), apoptosis rate of PC-3 cells treated with 50 mg/L, 100 mg/L and 200 mg/L SSG was increased. The proportion was 6.91%, 10.52% and 16.50% respectively. Conclusion: SSG can significantly suppress the cell proliferation and induce the aooptosis in PC-3 cells.
