摘要
目的探讨补骨脂素(PSO)加长波紫外线(UVA)光化学疗法(PUVA)诱导人白血病细胞K562凋亡时对Fas表达的影响。方法以K562细胞为研究对象,以细胞凋亡率、电镜下细胞超微结构改变以及细胞Fas基因表达水平、蛋白表达水平为检测指标,观察中药补骨脂中提取的PSO加波长为360 nm的UVA对人白血病细胞诱导凋亡的作用以及部分作用途径,并采用多因素方差分析法进行统计学处理。结果(1)单纯PSO、单纯UVA照射及PUVA均可诱导K562细胞发生凋亡,PUVA的作用显著强于前两者。(2)电镜下观察经PUVA处理后的K562细胞超微结构,可见明显的凋亡形态学改变。(3)单纯PSO、单纯UVA照射及PUVA均可上调K562细胞Fas基因表达水平,PUVA的作用显著强于前两者。(4)单纯PSO、单纯UVA照射及PUVA均可上调K562细胞Fas蛋白表达水平,PUVA的作用显著强于前两者。结论(1)PUVA可诱导白血病细胞K562发生凋亡,作用强于单纯PSO及单纯UVA照射。(2)PUVA诱导K562凋亡的途径之一为上调K562细胞Fas基因表达水平。
Objective To explore the effect of psoralen (PSO) and long-wave ultraviolet A (UVA) on apoptosis and expression of Fas in K562 leukemia cells. Methods K562 cells were taken as the study objects and their apoptosis ratios, ultrastructure changes and expression of Fas were detected in order that the effect of PSO extracted from Chinese herbal medicine and UVA (360 nm) on human leukemia cells could be observed. The factorial design and analysis of variance were used to analyze the interaction among the factors. Results ( 1 ) PSO, UVA and PSO + UVA (PUVA) all induced the apoptosis of K562 cells, but the effect of PUVA was stronger than that of the other two. ( 2 ) After having been treated with PUVA, ali the K562 cells showed obvious changes in ultrastructure related to apoptosis under electron microscope. (3) Although PSO, UVA and PUVA all increased the expression of Fas gene, the effect of PUVA was stronger than that of the other two. (4) PSO, UVA and PUVA all increased the expression of Fas protein, and the effect of PUVA was stronger than that of the other two. Conclusion PUVA can induce the apoptosis of K562 cells and its effect is the strongest. One of the pathways to induce apoptosis is to upregulate the expression of Fas gene.
