摘要
介绍了一种可准确、快速测定粮食中麦角甾醇含量的液相色谱方法。样品皂化后,用正己烷萃取麦角甾醇,不需要进一步纯化,直接上液相色谱分析。分析柱为Spherisorb硅胶柱,检测波长282nm。方法的检出限为0.1mg/kg,方法的回收率在95.1%~100.0%。分析的准确度评价采用微量制备技术和反相液相色谱分离技术,证明了该方法分析的结果准确可靠。
A sensitive,rapid ,reproducible and reliable liquid chromatographic method is described for determination of ergosterol in grain. The sample is saponified directly and the saponified mixture is extracted with n -hexane. Ergosterol is determined without further purification or clean up steps by using a liquid chromtagraph with a 250 × 4.0 mm column packed with spherisorb Si, 5μm, and a high pressure column prefilter. The ultraviolet detector is set at 282 nm. The limit of detection was 0.1 mL/m^3;recovery ranged 95.1% 100.0 %. The data analysed by this method is accurate, demonstrated by mini-preparative-HPLC method and reverse phase HPLC method.
