The Biologic Effects of the Immunosuppresive Agent Cyclosporine A on Mice Bone Metabolism in Vitro

摘要 目的探讨实验条件下免疫抑制剂环胞素A(CsA)在骨代谢过程中对破骨细胞和成骨细胞克隆的生物作用及其机制。方法用不同浓度的环胞素A与鼠骨髓细胞共同培养,诱导产生破骨细胞和成骨细胞克隆。分别以抗酒石酸酸性磷酸酶(TRAP)染色检查破骨细胞,碱性磷酸酶(ALP)染色检查CFU-F克隆和Von’Kassol染色检查CFU-OB克隆;光镜下计数破骨细胞数、CFU-F和CFU-OB成骨细胞克隆数。结果实验条件下环胞素A可诱导鼠破骨细胞的数量显著增加(F=16.496,P<0.001),且破骨细胞的数量与环胞素A剂量呈线性正相关(r=0.579,P<0.05);成骨细胞克隆CFU-F和CFU-OB的数量变化与环胞素A无相关性(F=1.380,F=0.305,P>0.05)。结论环胞素A在实验条件下可诱导鼠破骨细胞数量的增加,但对成骨细胞克隆无显著影响,环胞素A对骨代谢的生物作用可能是通过上调破骨细胞系统从而导致生物体发生骨质疏松。 Objective To investigate the biologic effects of immunosuppressive cyclosporine A on the osteoclastic cells and osteoblastic colony during the mice bone metabolism in vitro. Methods Bone marrow cells of mice were treated with different concentration of cyclosporine A in vitro. The mice stromal cells were cultured and osteoclastic cells and osteoblastic colony of CFU-F and CFU-OB obtained. TRAP positive multinuclear cells were measured by tartrate-resistant acid phosphatase(TRAP) staining, CFU-F and CFU-OB were checked with alkaline phosphatase(ALP) staining and Von' Kassol staining methods respectively. Results The number of TRAP positive multinuclear cells increased significantly( F = 16. 496, P 〈 0. 001 ) and depending on the CsA doses( r = 0. 579, P 〈 0.05 ). The number of CFU-F and CFU-OB were not significantly changed ( F = 1. 380, F = 0. 305, P 〉 0.05 ). Conclusion CsA can induce the osteoclastic cells increasing and has no significant effects for the CFU-F and CFU-OB. One of the major function of CsA for bone metabolism may be upregulation of osteoclast system and induction of osteopenia in the mice.
