摘要
醇提中药关木通粉末,经旋转蒸发、真空干燥后得到粗提药物浸膏;在饱和NaHCO3溶液中溶解浸膏,静置分层后调节pH至3得到马兜铃总酸粗品;经硅胶G柱层析,洗脱液为氯仿∶乙酸乙酯∶苯(2∶1∶1),重结晶洗脱液得到黄色结晶体。反相高效液相色谱鉴定并测定结晶体中马兜铃酸A含量。结果显示黄色结晶体为马兜铃酸A单体,结晶体中马兜铃酸A的含量达99.2%,表明该提取方法可成功获得纯度较高的马兜铃酸A。
Extracted aristolochic con-acid by dehydrate alcohol as the solvent from 150 g manchurian dutchmanspipe stem powder,get crude extract after rotary evaporation and vacuum dehydration, and dissolved extractum with NaHCO3 saturated solution,adjusted the pH to 3 after delamination. Used silica gel G chromatographic colum to laminar analysis. The eluent was made up of chloroform : acetidin : methylbenzol(2:1 : 1). Then get flav-erystal 0. 389g after recrystallization, identified and assayed the content with R-HPLC. the result showed that the flav-crystal was aristolochic acid A monomer, and the content reached 99.2%. The methold of extraction could get high purity aristolochic acid A,and it was a high performance way.
