摘要 目的:评价干化学法测定血清中茶碱浓度的可行性。方法:应用全自动干化学分析仪对30例服用茶碱的患者血清进行测定,并与免疫荧光偏振法进行比较,同时对干化学测定法进行方法学评价。结果:茶碱浓度在5.0~200!g/mL范围内线性良好(r=0.9985)。日内RSD为3.31%~5.85%,日间RSD为4.51%~7.39%;回收率为95.75%~102.33%。与免疫荧光偏振法进行比较,有良好相关性(r=0.9617,P>0.05)。结论:应用本方法测定血清中茶碱浓度重复性好,准确度高,与免疫荧光偏振法的测定结果差异无显著性,且需血量少,操作简便、快速,适合急诊检测。 Objective To evaluate the feasibility of dry-chemistry method for detecting the concentration of theophyline in serum. Methods The serum from 30 patients receiving theophyline therapy was detected using a fully automated dry-chemistry analyzer and the outcomes were compared with those determined by fluorescence polarization immunoassay. Results The linear range of the dry-chemistry method was 5.0 ~200 μg/mL (r=0.998 5) ;within and between-run CV were 3.31% ~5.85% and 4,51% ~ 7.39%,respectively; recovery rate was 95.75% ~ 102.33%. Drychemistry method was correlated with fluorescence polarization immunoassay (r=0.961 7 ) and the results measured by the two methods had no statistic difference (P 〉 0.05). Conclusion Automatic dry chemistry is a fast, accurate, and stable method for theophyline measurement, which is suitable for urgent detection.
