摘要
目的:研究大孔吸附树脂分离纯化槐角中总黄酮的工艺条件及参数,使分离工艺达到最优化。方法:以槐角总黄酮的吸附量和解吸率为指标筛选大孔吸附树脂,用紫外分光光度法测定槐角总黄酮含量对所筛选树脂分离纯化槐角总黄酮工艺进行评价。结果:D101型大孔树脂对槐角总黄酮的吸附性能和解吸性能较好,静态饱和吸附量为67.78mg.g-1,解吸率为97.58%;其吸附纯化条件为槐角提取物上样浓度为5.0mg.g-1,吸附流速为2.0BV.h-1(树脂床体积),以70%乙醇作为洗脱剂进行洗脱效果最佳。经D101树脂纯化后槐角总黄酮的含量为67.3%。结论:该法简单可行,分离效果好,能满足于大生产的要求。
objective: To optimize the purification method of the total flavones in Huaijiao by macroporous resin. Methods: The static adsorption/desorption characteristics were taken as index to select the macroporous resin. The separating efficiency was evaluated by measuring the concentration of total flavones in Huaijiao with UV spectrophotometer. Results: The D101 macroporous resin had the best separating efciency. The static adsorption capacity/desorption ratio of the total flavones were 67.78mg·g^-1/97.585 %. When the concentration of the Huaijiao extract sample reached 5.0mg/mL, the current velocity of adsorption was 2.0BV. h-1 (resin bed volume), and 70% ethanol as eluant, the purity of total flavones Could reach 60%. Conclusion: The method is simple and feasible, with good separating effect which can meet industrial requirements.
