摘要
[目的]寻求金线莲原球茎诱导、增殖、分化与生根的最佳培养基及影响原球茎增殖与分化的主要因子。[方法]以金线莲茎段为外植体,MS为基本培养基,附加不同浓度的6-BA、NAAT、DZ和S-3307,对原球茎的诱导、继代增殖、苗的分化、生根等进行研究。[结果]原球茎诱导的最佳培养基为MS+TDZ 0.4 mg/L+NAA 0.2 mg/L,诱导率达93.3%;原球茎增殖的最佳培养基为MS+6-BA 2.0 mg/L+S-33071.0 mg/L+NAA 0.2 mg/L,增殖系数达9.4;芽分化的最佳培养基为MS+S-33071.5 mg/L+TDZ 0.4 mg/L+NAA 1.5 mg/L+琼脂0.7%,分化率达91.7%;最佳的生根培养基为1/2 MS+NAA 0.2 mg/L+活性炭0.05%+琼脂0.7%,生根系数达4.4;组培苗移栽30 d后,存活率均达93%左右。[结论]S-3307是金线莲原球茎增殖的主要因子,TDZ是原球茎分化的主要因子。
[Objective] The research aimed to seek the optimum medium for the protocorm induction, proliferation, differentiation and rooting in Anoectochilus roxburghii and main factors that affected the protocorm proliferation and differentiation. [Method] With the stem segments of A. roxburghii as explants, 6-BA, NAA, TDZ and S-3307 with different concn, was appended on basic MS medium to study the protoeorm induction, subculture proliferation, the differentiation of seedlings, rooting and so on. [Result] The best medium for the pmtocorm introduction was MS+ TDZ 0.4 mg/L + NAA 0.2 mg/L with the induction rate of 93.3%. The best medium for the protocorm proliferation was MS + 6-BA 2.0 mg/L + S-3307 1.0 mg/L + NAA 0.2 mg/L with the multiplication coefficient of 9.4. The best medium for bud differentiation was MS + S-3307 1.5 mg/L + TDZ 0.4 mg/L + NAA 1.5 mg/L + agar 0.7%, with the differentiation rate of 91.7 %. The optimum rooting medium was 1/2 MS + NAA 0.2 mg/L + activated cart)on 0.05 % + agar 0.7 % with the rooting coefficient of 4.4. After the tissue culture seedlings were transplanted for 30 d, the survival rate reached about 93%. [Conclusion] S-3307 was main factor for protocorm proliferation of A. roxburghii and TDZ was main factor for protocorm differentiation.
