摘要
[目的]为探索暗纹东方基因组中是否存在BAFF基因。[方法]通过比对人、小家鼠、野猪、原鸡的BAFF基因cDNA序列,获取高度保守的同源片断。根据红鳍东方基因组文库序列设计引物,从野生暗纹东方肌肉样品中提取总DNA进行PCR扩增,并进行PCR产物的克隆与测序。[结果]通过PCR扩增获得3条特异性条带,大小分别为542、349、221 bp。在红鳍东方基因组文库中未找到与542 bp序列匹配的片段。349和221 bp序列在暗纹东方和红鳍东方中是保守的。暗纹东方与红鳍东方序列比对结果表明,碱基突变的主要方式为插入或缺失。[结论]3个扩增条带为BAFF基因相关序列的可能性较小。在鱼类生物体中很可能不存在与哺乳类、鸟类BAFF同源基因。
[ Objective] The research aimed to explore whether there was BAFF gene in the genome of Takifugu obscurus. [ Method] eDNA sequences of BAFF gene from Homo sapiens, Mus musculns, Sns scrof and Ga/lns gallus were compared to obtain its highly-conserved homologous fragment. Primers were designed according to the sequences in the genomie library of Takifugu rubripes . Total DNA was extracted from the muscle samples of wild T. obscures to make PER amplification and the PER products were cloned and sequenced. [Result] Three specific bands were obtained though PCR amplification, with the size of 542, 349, 221 bp. The suited fragment with 542 bp sequence was not found in the genomie library of T. rubripes. Sequences with the size of 349 and 221 bp were conservative in T. obscurus and T. rubripes . The comparison results between T. obscurus and T. rubripes showed that the main type of base mutation was insertion or deletion. [Conclusion] The possibility that 3 amplified bands were the related sequences to BAFF gene was less. It was probably that the homologous gene with BAFF of mammalian and birds didn' t exist in fish organism.
