单细胞凝胶电泳技术检测大鼠脑缺血再灌注后DNA损伤及海风藤提取物的干预作用

摘要 目的用单细胞凝胶电泳技术(SCGE)检测局灶性脑缺血再灌注(I/R)大鼠神经细胞DNA损伤及海风藤干预作用。方法应用线栓法建立Wistar大鼠大脑中动脉阻塞(MCAO)再灌注模型,单细胞凝胶实验技术分别观察假手术组、模型组、二甲基亚砜(DMSO)组、海风藤提取物治疗组缺血灶周神经细胞DNA损伤的变化。结果与假手术组比较,模型组和DMSO组DNA损伤率6~72h明显增加,但海风藤组DNA损伤率与其他组比较24、72h有明显下降(P<0.01,P<0.05)。结论海风藤可以减轻I/R后神经细胞DNA损伤,对缺血脑组织有保护作用。 Objective To investigate DNA damage after focal cerebral ischemia-reperfusion and intervention of extraction from Piper kadsura Ohwi by single cell gel electrophoresis (SCGE). Methods The models of middle cerebral artery occlusion (MCAO) and reperfusion were established to detect DNA damage of nerve cell in the cerebral tissue among sham operated, model, DMSO-treated ( dimethyl sulfoxide) and extraction from Piper Kadsura Ohwi-treated groups. Results Comparing shame operated group, model and DMSO groups had the obvious increase in DNA dagamge rate during 6 - 72 h, but Piper Kadsura Ohwi group had significant decrease during 24 -72 h( P 〈 0.01 , P 〈 0.05 ) compared with the other groups. Conclusions Extraction from Piper kadsura Ohwi plays a significant role in protecting cerebral tissue through decreasing the DNA damage caused by isehemia-reperfusion.
