摘要 目的探讨FK506对人黑素瘤细胞增殖及黑素合成的影响。方法体外人A375黑素瘤细胞培养,MTT法测定FK506对黑素瘤细胞增殖的影响,NaOH法测定FK506对黑素合成的影响。结果浓度为10^1~10^2 nmol/L的FK506对黑素瘤细胞增殖无影响,10^3~10^4 nmol/L的FK506对黑素瘤细胞增殖有抑制作用,但是10^1~10^4 nmol/L的FK506对黑素瘤细胞的黑素合成均有促进作用,其中以10^1~10^3 nmol/L的FK506对黑素合成的促进作用最为显著。结论FK506能够直接促进培养的黑素瘤细胞的黑素合成。 Objective To study the effects of FK506 on cell proliferation and melanin synthesis of human melanoma cells, Methods Human A375 melanoma cells were cultured and treated with various concentration of FK506. Cell proliferation was measured with MTT assay, The synthesis of melanin was determined by NAOH assay, Results FK506 of 10^1- 10^2 nmol/L did not influence the proliferation of melanoma cells. Although FK506 of 10^3 - 10^4 nmol/L could inhibit the proliferation of melanoma cells, FK506 of 10^1 - 10^4 nmol/L could all promote the melanin synthesis of melanocytes, The significant concentration of FK506 ranged from 10^1 to 10^3 nmol/L, Conclusion FK506 could promote the melanin synthesis of melanoma cells,
