摘要
目的:观察C5a、PMA、A23187、LPS对PBMCIL-8产生的影响,为研究减轻C5a致病作用的途径创造条仵。方法:用rhC5a、PMA、A23187、LPS诱导PBMC,以ELISA法测定细胞培养上清中IL-8。结果:PBMC经rhC5a、PMA、A23187和LPS诱导后IL-8分泌水平较对照组增高。结论:激活PKC、提高细胞内钙离子浓度可使IL-8分泌增加,提示用PKC抑制剂、钙通道拮抗剂有可能使C5a介导的PBMCIL-8分泌水平降低。
Objective:Interleukin-8(IL-8) from human peripheral blood mononuclear cells(PBMCs) and stimulated with C5a,PMA,A 23187 and LPS was examinated for finding a way to reduce the pathogensis of C5a.Method:The IL-8 in supernatant of PBMCs which were induced with rhC5a,PMA,A 23187 and LPS was detected by ELISA.Result:The secretion of IL-8 from PBMCs induced with rhC5a,PMA,A 23187 and LPS increased in comparison with the control group.Conclusion:Promotion of PKC or cytoplasma calcium increased IL-8 secretion and PKC inhibitor or calcium channel antagonist decreased IL-8 secretion from PBMCs induced with C5a.
