摘要
制备硫酸软骨素(ChS)铜配合物,分析其光谱学和热力学特征,再将ChS进行羧基还原、脱硫和硫酸酯化等化学修饰制备成相应衍生物,用荧光淬灭实验和平衡透析实验分析ChS及衍生物与铜离子的配位特性。ChS铜配合物的光谱学和热力学性质变化说明ChS与铜离子发生了配位作用。荧光淬灭实验和平衡透析实验结果显示:将ChS进行羧基还原或多硫酸化后,ChS与铜离子的配位作用减弱,而脱硫后的ChS的配位作用增强,说明是ChS中的羧基而非硫酸基与铜离子发生了配位作用。
This experiment studied the ChS-Cu (chondroitin sulfate-Cu) complex, analyzed its speetroscopic and thermodynamics characteristics, and its three chemically reduced or sulfonated or desulfate derivatives, and compared the binding properties of unmodified ChS and its three chemically reduced or sulfonated or desulfate derivatives by fluorescence quenching assay and equilibrium dialysis technique. The changes of spectroscopic assay and thermodynamics of ChS-Cu complex exhibit the mating reactions between ChS and Cu^2+. The results of luorescence quenching assay and equilibrium dialysis experiments showed that the carboxy group but not sulfate group is involved in this reaction.
