反相高效液相色谱法(RP-HPLC)测定异构颗粒酒花中的异α-酸 被引量:6

摘要 利用反相高效液相色谱法(RP-HPLC)研究异构颗粒酒花中异α-酸的测定方法。结果表明:当检测波长270nm、流动相为甲醇:水:磷酸(85%):EDTA(ethylene diamine tetra aceticacid)二钠盐(0.2mol/L)(770:210:5:1)、流速为1.0ml/min时,方法的精密度为1.09%,外加标回收率91.4%~97.1%之间。RP-HPLC的色谱柱为EC250/4Nuclesoil,100-5C18(4.0×250nm,5μm),保护柱为CC8/4Nuclesoil100-5C18。该方法可以简便、准确地测定异构颗粒酒花中异α-酸含量。 Iso- α -acids content of isomerized hop pellets was analyzed by reversed-phase high performance liquid chromatography (RP-HPLC). The optimized conditions are: 270 nm by ultraviolet detection with methanol: H2O:H2PO4 (85%): EDTA- Na2 (0.2 mol/L) (770:210: 5: 1, respectively) as mobile phase with flow rate 1.0 ml/min. The average recovery of iso- α -acids is 91.4 %-97.1%, and the accuracy of the method as RSD (relative standard deviation) is about 1.09 %. The column is EC 250/ 4 Nuclesoil, 100-5 C18 (4.0 × 250 nm, 5μm) with Guard Cartridge CC 8/4 Nuclesoil 100-5 C18. It is a convenient and accurate method for analyzing iso-α-acids content in isomerized hop pellets.
