血管内皮生长因子和地塞米松对舌鳞状细胞癌细胞株基质金属蛋白酶-9表达的影响

摘要 目的探讨血管内皮生长因子(vascular endothelial growthfactor,VEGF)和地塞米松(dexamethasone,DEX)对舌鳞状细胞癌(鳞癌)细胞株基质金属蛋白酶(matrix metalloproteinase,MMP)-9表达的影响。方法对体外培养的舌鳞癌细胞株分别用VEGF和DEX作为刺激因子,采用免疫组化SP法测定癌细胞中MMP-9蛋白含量的变化。结果与对照组相比不同浓度的VEGF(1、5、10 ng/ml)作用于舌鳞癌细胞24 h后,MMP-9的活性差异有显著性(P<0.01),其与VEGF有剂量依赖关系;加入DEX(0.05%)后,MMP-9表达降低,与未加入DEX的VEGF组相比差异有显著性(P<0.01)。结论VEGF可以诱导舌鳞癌细胞MMP-9的表达;DEX可以抑制舌鳞癌细胞MMP-9的表达。 Objective To investigate the effects of matrix metalloproteinase-9 in tongue squamous cell line induced by VEGF and DEX. Methods The effects of VEGF and DEX on the production of MMP-9 in tongue squamous cell were studied by immunohistochemistry. Results VEGF had a significantly different effect on the activity of MMP-9 in tongue squamous cell line after 24h, and the activity of MMP-9 was more dependent on VEGF. The activity of MMP-9 may be decreased by the effects of VEGF and DEX. The activity of MMP-9 was also dependent on VEGF. Conclusion VEGF may promote the production of MMP-9 in tongue squamous cell;DEX may decrease the production of MMP-9 in tongue squamous cell.
