新型血小板添加液对血小板低温液态保存效果的实验研究 被引量:2

摘要 目的探讨使用改良的血小板添加液(PAS)替代70%自体血浆在低温液态条件下对血小板的保存效果。方法采集6单位单采血小板,每一单位单采血小板平均分为3组,A组加入70%PAS-ⅢM/30%血浆10℃保存;B组加入100%血浆22℃保存;C组加入100%血浆和海藻糖-85℃保存。A、B组在第1、5、7、9天取样检测,C组在20d后复苏取样检测。分别检测PLT计数、PDW、MPV、CD62p、HSR、PAgT、pH值、LDH以及GLU的变化。结果保存期内随保存时间延长,A组和B组CD62p表达增加,HSR和最大聚集率降低;A组的LDH释放、GLU消耗、CD62p表达、HSR、血小板最大聚集率与B组比较,差异有统计学意义(P<0.05)。B组pH值在第5天起明显降低(P<0.05)。C组LDH释放明显较其他两组增多,但CD62p表达较少(P<0.05)。C组PDW显著升高,除与B组第5天结果相似外,均高于A、B组各时间点结果(P<0.05)。各组PLT计数相差不显著。结论改良的PAS-ⅢM能够很好的替代血浆用于血小板的保存,同时在低温条件下能够很好地保护血小板的功能,保存效果好于血浆常温保存。 Objective To explore the preservation effect of a modified platelets (PLT) additive solution ( PAS- ⅢM) as a substitute for plasma in the storage of platelet concentrates at low temperature. Methods Apheresis platelets from 6 donors were divided and stored in different media, that is, A:70% PAS- Ⅲ M/30% plasma stored at 10 ℃ , B : plasma stored at 22 ℃, C : plasma/trehalose stored at - 85 ℃. The samples of in group A and B were respectively collected in 0, 1,5, 7 and 9 days after storage, and those of group C on day 20 after storage. Platelet count (PLT), mean platelet volume (MPV), platelet distributing width (PDW), CD62p, hypotonic shock response(HSR), platelet aggregation (PAgT), pH, lactic dehydrogenase (LDH), glucose consumption were evaluated. Results The expression of CD62p in group A and B group were increased in a time-dependent manner, but HSR and PAgT were decreased. CD62p expression, LDH release, glucose consumption were significant higher, and HSR, PAgT were significant lower in group B than in group A ( P 〈 0.05 ). After 5 days' storage, pH value in group B were significant decreased ( P 〈 0.05 ). LDH release were significant higher in samples of C group and the expression of CD62p were lower than those of other 2 groups ( P 〈 0.05 ). PDW of C group were increased after storage and significant higher than that of other 2 groups ( besides that of B group on day 5 ) ( P 〈 0.05 ). In all storage groups, PLT had no significant difference. Conclusion PAS- Ⅲ M could be a good substitute for plasma in platelet storage, and can protect the platelets function at low temperature.
