摘要 目的为探讨低功率毫米波对HL60细胞活力影响的机制。方法应用细胞培养、活细胞计数、光镜、电镜及增殖细胞核抗原(Proliferating cell nuclear antigen,PCNA)免疫组化染色及图像分析等方法,检测低功率(功率密度约2mW/cm2)及频率41.32GHz的毫米波辐照HL60白血病细胞增殖的影响;分为辐照时间15、30、45、60min组和对照组(0min组)共5组。结果与对照组比较,毫米波辐照45min和60min组,HL60细胞活性增加和PCNA阳性细胞增多,光镜观察各组未见明显差别,超微结构显示HL60细胞形态呈增殖改变。结论低功率毫米波辐照对HL60细胞有促进增殖的作用。 Objective To investigate the mechanism on the irradiation effect of low power millimeter waves (MMW) to HL60 leukemia cells. Methods HL60 cells, irradiated by millimeter wave at frequency of 41.32 GHz (mean power density 〈2 mW/cm^2) , were divided into 5 groups according to the irradiation time (0, 15, 30, 45, 60 min groups), The irradiation effect to cell proliferation was detected by cell counting, proliferating cell nuclear antigen (PCNA) immunohistoehemieal staining and quantitative analysis. Light microscopy and electron microscopy were used to observe the morphological changes of irradiated cells. Results The proliferation activity and number of PCNA-positive cells were increased in HL60 cell after 45 and 60 min MMW irradiation. There was no difference in the morphology between HL-60 cells exposed to MMW and sham-exposed group. Ultrastructure changes were consistent with cell proliferation. Conclusion Low power MMW irradiation may promote HL60 cell proliferation.
