高效稳定表达人缺氧诱导因子抑制因子肠上皮细胞株的建立 被引量:1

摘要 目的通过转染含有人FIH全长cDNA的质粒pcDNA3.1-V5-His-A-FIH,建立FIH高效表达的肠上皮细胞株。方法对pcDNA3.1-V5-His-A-FIH进行扩增、提取,用脂质体Lipofectamine 2000将pcDNA3.1-V5-His-A-FIH转入Caco-2细胞,经G418筛选并逐步传代,RT-PCR和Western blot法检测阳性细胞克隆FIH mRNA和蛋白表达。结果转染pcDNA3.1-V5-His-A-FIH后Caco-2细胞FIH mRNA是普通Caco-2细胞的1.98倍,而FIH蛋白含量是普通Caco-2细胞的1.70倍。结论成功建立了高效、稳定表达人FIH基因的肠上皮细胞株。
