摘要
目的获得鸡含缬酪肽蛋白(Valosin-containing protein,VCP)的cDNA和氨基酸序列,为进一步研究VCP和视网膜发育及病变的关系提供理论依据。方法根据已发表的鸡ATP酶p97的基因序列片段设计引物,从白来杭鸡视网膜cDNA文库中获得VCP全基因序列,并进行序列分析和比较。结果获得了VCP的全长cDNA序列,此基因cD-NA全长为有2 916 bp,并有2 421 bp的完整开放阅读框,编码806个氨基酸。鸡VCP的基因及其对应的氨基酸序列与人、小鼠VCP基因及其对应的氨基酸序列高度同源,并具有ATP酶结合位点的高度保守AAA序列。结论获得了全长cDNA序列已在Genbank中登录(AB195711),为从基因及蛋白质水平研究VCP和视网膜发育及病变的关系奠定了基础。
Objective Cloning and structure analysis of Valosin-containing protein(VCP) cDNA.Methods Based on the amino acid sequence of having known the fraction of chick ATPase p97 primers were designed.The cDNA of VCP was obtained from a chick newborn retinal cDNA library,and cDNA was used for database analysis.Results A cDNA clone with a length of 2 916 base pairs was isolated.The sequence analysis showed that the cDNA contains an open reading frame with 2 421 bp encoding a 806 amino acid protein.Analysis of public databases revealed that the molecule showed the highest homology to human and mouse VCP.VCP is a member of AAA family proteins and share the conserved ATP binding domain.Conclusion VCP has been deposited in GenBank under accession number AB195711.The present results would be valuable in analyzing function of VCP at both nucleotide and protein levels.
