人静脉移植物eNOS和P-eNOS的表达

摘要 目的检测人静脉移植物中eNOS的表达及其活性。方法应用免疫荧光组织化学技术对30个静脉移植物再狭窄标本中eNOS的表达进行了检测,用抗磷酸化的eNOS抗体(P-eNOS)检测eNOS的活性。用激光共聚焦显微镜拍片,图片用Silicon Graphics Octane进行处理。结果在正常静脉血管中,eNOS在内皮细胞不表达;在病变静脉血管,在内膜增生早期,eNOS在管腔面内皮细胞的表达明显上调,在外膜小血管的表达也增加,随着内膜增生的不断发展,eNOS在管腔面内皮细胞的表达明显下降,外膜小血管逐渐从外膜向中膜延伸。免疫双重染色证实eNOS和p-eNOS的表达共同定位于内皮细胞。结论eNOS在静脉移植物中的表达模式提示其可能参与了中膜毛细血管的生成以及新内膜的形成。 [Objective] To detect expression of eNOS and its activity in human vein graft. [Method] Thirty samples of human vein grafts were used for this study expression of eNOS and its activity by using confocal immunofluorescence with specific antibodies against eNOS and phosphorated eNOS (P-eNOS). The data were processed with silicon graphics octane system. [Results] In normal veins, eNOS was not expressed in the endothelial cells; In diseased vein grafts, eNOS was significantly induced in the luminal surface of endothelial cells at early stage of intima formation. Positive staining of eNOS was also observed in the adventitia. In the vein grafts with thick intima, eNOS expression in luminal surface of endothelial cells was decreased. In the media eNOS expressed was located in the capillaries. Double immunostaining demonstrated the co-localization of of eNOS with P-eNOS in endothelial cells. [Conclusion] The pattern of eNOS expression implies its involvement in neointima formation and tunica media angigenesis.
