摘要
目的:检测TNF-α作用前后人乳腺癌细胞株MCF-7中CD44s、CD44v3和CD44v6的表达水平的变化,及对细胞侵袭能力的影响。方法:采用RT-PCR和Westernblot方法检测TNF-α作用前后人乳腺癌细胞株MCF-7中CD44s、CD44v3和CD44v6的表达情况,了解TNF-α对CD44各亚型表达水平的影响;Transwell检测TNF-α作用前后人乳腺癌细胞株MCF-7侵袭力的变化情况。结果:TNF-α作用后,MCF-7细胞株CD44smRNA表达下降,CD44v3mRNA和CD44v6mRNA的表达上升。相应蛋白表达与mRNA水平呈现一致性改变,其中CD44s蛋白表达下降,CD44v3蛋白和CD44v6蛋白表达上升;TNF-α作用后细胞侵袭力提高(P<0.001)。结论:在人乳腺癌细胞株MCF-7中,TNF-α可能通过影响CD44各亚型mRNA和蛋白表达水平,进而促进肿瘤细胞的侵袭能力。
Objective:To detect the expression of CD4s, CD4v3 and CD4v6 in human breast cancer cell line MCF-7 cells treated by TNF-α, analyzing the relationship between CD44 expression and TNF-α and study the effect on the invasion of breast cancer cell line MCF-7. Methods : MCF-7 were treated by TNF-α for 24h and 48h, respectively, and observed of the proliferation by MTT. The mRNA and protein expression of CD44 were analyzed in human breast cancer cell line MCF-7 cells after TNF-α treatment for 24h by RT-PCR and Western blot assay. The relationship between CD44 expression and TNF-α was investigated. An model of cancer cells was made by the Boyden chamber and the artificial matrix membrane in vitro. The number of the cells through the artificial matrix membranein every group was counted to compare the change of the invasion capability among the expermental groups. Results:TNF-α treatment reduced the level of CD4s mRNA and protein by 16%-38% and 5% -47%, respectively. Meanwhile TNF-α treatment increased the level of CD4v3 mRNA and protein by 5%-52% and 2%-79%, respectively. At the same time, TNF-α induced the express of CIM4v6 mRNA and protein by 2% -45% and 4%-35%, respectively. Conclusion: TNF-α modulate the express of CD44 in human breast cancer cell line MCF-7 cells, affecting their migration and invasion in vitro.
