免疫磁珠联合RT-PCR检测胃癌肿瘤标志物α4GnT

目的:研究α4GnT mRNA的转录本在胃癌患者和消化性溃疡患者外周血中的表达,并分析其在胃癌及消化性溃疡两种疾病间的差别及与胃癌预后的关系。方法:免疫磁珠分选25例胃癌患者及10例消化性溃疡患者外周血中上皮来源的细胞联合RT-PCR法检测α4GnT mRNA的转录本。结果:α4GnT mRNA在胃癌与消化性溃疡患者中阳性率分别60%(15/25)和40%(4/10),两者之间的差异无显著性(P>0.05)。α4GnT mRNA在早期胃癌与进展期胃癌患者中阳性率分别17%(1/6)和79%(15/19),两者之间的差异有显著性(P<0.05)。结论:运用免疫磁珠联合RT-PCR检测外周血肿瘤标志物α4GnT在鉴别胃癌与消化性溃疡方面无意义,但可作为预测胃癌预后的指标之一。

Objective： To study the expression of α4GnT mRNA and to analyze the difference between gastric cancer and peptic ulcer and the relationship with prognosis in gastric cancer. Methods： Immunizations magnetic separate epithelial cells from peripheral blood of 25 patients with gastric cancer and 10 patients with peptic ulcer and combined RT-PCR detecting the transcripts of α4GnT. Results ：There showed that the positive rate of α4GnT mRNA in gastric cancer and peptic ulcer was 60% （ 15/25 ） and 40% （4/10）, respectively, there was no significant difference（ P 〉 0. 05 ）. The positive rate of α4GnT mRNA in early gastric cancer and progressive gastric cancer was 17% （1/6） and 79% （ 15/19 ）, respectively, the difference was significant （ P 〈 0. 05 ）. Conclusion.. There is no significance to detect tumor marker α4GnT from peripheral blood using immunizations magnetic combined with RT-PCR in identification of gastric cancer patients and peptic ulcer patients, but it can be used as one of the factors to fore fast the gastric cancer prognosis.
