摘要 目的观察氯化血红素(hemin)对大鼠肝脏血红素加氧酶-1(HO-1)的诱导表达作用。方法SD大鼠18只随机分为两组:氯化血红素处理组共10只,每日早晨hemin 15mg/kg腹腔注射,每24h重复1次;生理盐水对照组共8只,每日早晨同时间腹腔注射等量生理盐水,每24h重复1次。第10d处死大鼠后检测血清ALT、AST水平,用免疫组织化学方法观察肝组织HO-1的表达,采用逆转录聚合酶链反应(RT-PCR)检测肝组织HO-1mRNA表达。结果hemin处理组与生理盐水对照组大鼠血清ALT,AST水平无明显差异。hemin处理组肝组织HO-1表达明显高于生理盐水对照组。结论hemin能诱导大鼠肝组织HO-1表达明显增加。 Objective To evaluate the expression level of heme oxygenase - 1 in rats liver induced by hemin. Methods 18 SD rats were divided randomly into observation group and control group each consisted of 10 rats. -The rats in observation group treated with hemin were injected hemin intraperitoneally (15mg/kg) every morning. The 8 rats of control group treated with saline were injected saline intraperitoneally. On the 10th day of treatment all rats were killed and the levels of plasma ALT, AST were determined. Immunohistoehemical staining were employed to evaluate the expression of heme oxygenase - 1. Reverse transcription - polymerase chain reaction ( RT - PCR ) were used to examine the expression of HO - lmRNA in the liver. Results The levels of plasma ALT and AST were no significant difference between the group treated by hemin and the control group treated by Saline. The expression level of heme oxygenase - 1 in liver of the treatment group was more significant than that the control group. Conclusion Hemin can induce high expression of heme oxygenase - 1 in rats liver.
