摘要
目的:研究抗血管内皮细胞生长因子抗体对视网膜新生血管化的抑制作用。方法:以75%氧气和25%氮气的高浓度氧诱导C57BL/6J小鼠建立缺血性视网膜病变模型,应用不同浓度抗血管内皮细胞生长因子抗体进行玻璃体腔内注射,在组织切片上计数和比较正常和实验条件下以及不同浓度药物注射后视网膜新生血管芽细胞核数。结果:缺血性视网膜病变小鼠模型有较多的视网膜新生血管芽细胞核数,而玻璃体内注射抗血管内皮细胞生长因子抗体的小鼠视网膜新生血管芽细胞核数明显减少(两组用药组与高氧对照组间P均<0.01),特别是大剂量用药组减少54.61%。结论:抗血管内皮细胞生长因子抗体能有效抑制视网膜的新生血管化。
Objective:To evaluate the effect of suppression of ischemia-induced retinal neovascularization in vivo by inhibition of vascular endothelial growth factor(VEGF). Method:Murine models of hyperoxia-induced ischemic retinopathy were established. Intravitreal injections were performed with anti-VEGF monoclonal antibody or NS. The nuclei of new vessel buds extending from the retina into the vitreous in different groups are counted and compared. Results:There were plenty of new vessel buds in the eyes of mice in hyperoxia condition. Retinal neovascularization in the eye injected with large-dosage anti-VEGF monoclonal antibody was reduced 54.61% compared with that of the hyperoxia control group (P〈0.01). Conclusion:The retinal neovascularization were suppressed by using anti-VEGF monoclonal antibody.
