四氧化锇固定液长期保存的探讨

Long-term preservation of osmic acid fixative solution

目的:探究一种可行性高、方法简便易行的四氧化锇固定液长期保存方法。方法:将四氧化锇固定液(1%)分组冷冻,设3个实验周期组,分别是,6月组,12月组,24月组。结果:与对照组比较,各实验组四氧化锇固定液冷冻后其工作效能相同,电镜(TEM)照片显示,组织超微结构同样清晰。结论:四氧化锇固定液可以冷冻长期保存,冷冻后其工作效能不变。

Objective.To explore a highly feasible and easy method for the long--term preservation of osmie acid fixative solution. Methods ：Osmic acid fixative solution （1%） was frozen at freezer （general size in lab） in 3 groups,which were divided into 6-- month group, 12--month group and 24--month group, respectively. Results ：The electronmicroscope pictures from the 6-- month group, 12-- month group and 24 --month group showed that the tissue ultrastrueture was as clear as the pictures from the control group. Conclusion. The Osmic acid fixative solution may be frozen for long--term preservation, with no change in its working potency and functions after freezing.
